Cu(II)-KOMPLEXEK éS Fe(II)-VEGYüLETEK KöLCSöNHATáSAINAK
VIZSGáLATA BIOKéMIAI SZEMPONTBóL

Dolgozatom rávilágít azokra a jelenségekre, amelyeket az általam vizsgált réz(II)-vegyületek és Fe(II)-sók kölcsönhatása során figyeltem meg. A dolgozat bevezetője bemutatja a rézvegyületek vizsgálatának biokémiai fontosságát, pontosabban ezek szerepét a reaktív oxigén intermedierek képződésében. Bemutatja azt a sokak által leírt jelenséget, miszerint egyes antioxidánsok jelenlétében a rézvegyületek in vivo szabadgyököket fejlesztenek.
Az „Anyag és módszer” fejezet bemutatja az általunk vizsgált rendszert, amely Cu(II)-illetve Cu(I)-citrátot, vas(II)-szulfátot illetve redukálószereket tartalmaz, valamint leírja a rendszer illetve a keletkezett anyagok tanulmányozási módszereit: spektrofotometriás kinetikai meghatározások, az elkülöníthető termékek jodometriás réztartalom-meghatározását, a reaktív oxigén intermedierek mennyiségi meghatározását, valamint a rendszerben található Cu(I)-citrát tanulmányozását acetonnal történő komplexképzés alapján. Az „Eredmények” fejezet bemutatja az egyes meghatározások által nyert számadatokat. A „Megbeszélés” fejezetben az „Eredmények” alapján igazolva van, hogy mind a Cu(II)-, mind a Cu(I)-citrát oldható Fe(II)-só hatására kémiai átalakuláson megy keresztül, mely abban nyilvánul meg, hogy a Cu(II)-citrát réz-dioxigén-komplexek képződésére válik alkalmassá, a Cu(I)-citrátnak pedig az acetonnal való reakciókészsége megváltozik. Ugyanitt be vannak mutatva azok az egyensúlyok, melyek az átalakulás alatt álló rendszerben lejátszódnak a különböző Cu(I)- illetve Cu(II)-citrátok közt. A „kék oxidázok” aktív centrumának működéséhez van hasonlítva egy folyamat, mely során a rendszerben egy zöld színű, bicentrikus m-oxo-komplex rézsója képződik. A „Következtetések” fejezet részletezi a kapott eredmények kémiai és biokémiai értelmét, a rendszer működése alapján körvonalazódhat egy lehetséges mechanizmus, mely szerint megérthető a bevezetőben tárgyalt paradoxális kölcsönhatás Cu(II) és antioxidánsok között.

THE STUDY OF THE INTERACTION BETWEEN Cu(II)-COMPLEXES AND
Fe(II)-COMPOUNDS FROM THE ASPECT OF BIOCHEMISTRY

This work presents interactions that the author managed to describe by studying the interaction between the Cu(II)-complexes and Fe(II)-salts. The introduction relates the biochemical importance of the investigation of compounds containing copper, their role in the development of reactive oxygen substances. It presents the production of free radicals in the presence of copper-complexes and antioxidants, a fact stated by several authors.
The chapter entitled “Material and methods” presents the studied system which consists of Cu(II)- and Cu(I)-citrate, ferrous sulphate and reductive agents, it describes the methods of investigation: spectrophotometrical kinetical experiments, the determination of the composition of some isolable compounds by iodometry, the quantitative study of the reactive oxygen substances formed in the system and the study of formed and previously added Cu(I)-citrate by complexation with acetone. “Results” contains the numerical data of the experiments. “Discussion”, based on results of the experiments demonstrates that both Cu(I)- and Cu(II)-complexes underwent a chemical change which probably consists of the alteration of the coordinative geometry of the copper centre. This change can be observed by the capability of the newly formed Cu(II)-citrate to form a copper-dioxygen adduct . The reactivity of Cu(I)-citrate towards acetone is changed too. Equilibria taking place between previously added and newly formed Cu(I)- and Cu(II)-citrates are also presented. A reaction which results in forming of an insoluble green compound, -the Cu(II)-salt of a dicentric oxo-copper complex- is compared with a biochemical reaction, the function of the catalitical centre of the blue oxidases. “Conclusions” presents the chemical and biochemical importance of the obtained results.
Having carried out the study of the system, possible mechanism have been found by which the paradoxal interaction between copper salts and antioxidants, described in the “Introduction” can be understood.
